whats about that area? where does that duct feed from? if that duct has a slight grade on it, water from another area could have traveled along it and fell onto the dry wall . Or if there is a bathroom above there (which it doesnt look like there is) you can have a leek and not be seeing it or if there is a tub/shower above there ... excessive amounts of water spilling out of the shower can seep through the floor. just do some investigating
